Fluorescent Tyramide

Fluorescent tyramide is a signal-amplification reagent used to visualize low-abundance proteins or nucleic acid targets with high spatial precision. In tyramide signal amplification, horseradish peroxidase converts fluorescently labeled tyramide in the presence of hydrogen peroxide into reactive radicals that covalently attach to nearby proteins, depositing many fluorophores around the detection site. In genetics, this method strengthens signals in fluorescence in situ hybridization, immunofluorescence, and related assays for gene, transcript, and protein localization. Its localized deposition supports sensitive detection, sequential staining, and multiplex imaging, helping researchers analyze molecular expression patterns in cells and tissues.

High Resolution Fluorescent In Situ Hybridization in Drosophila Embryos and Tissues Using Tyramide Signal Amplification

The described RNA in situ hybridization protocol allows the detection of RNA in whole Drosophila embryos or dissected tissues. Using 96-well microtiter plates and tyramide signal amplification, transcripts can be detected at high resolution, sensitivity, and throughput, and at a relatively low cost.

Several different methods have been established for multiplex immunostaining using primary antibodies from the same host species. Here, we describe the use of microwave-mediated antibody stripping and of fluorophore-tyramide to block antibody cross-reactivity during multiplex immunostaining on formalin-fixed paraffin-embedded mouse adrenal sections.

Localizing gene expression to specific cell types can be challenging due to the lack of specific antibodies. Here we describe a protocol for simultaneous triple detection of gene expression by combining double fluorescence RNA in situ hybridization with immunostaining.
