Nuclei Permeabilization

Nuclei permeabilization is the controlled modification of the nuclear envelope to allow enzymes, antibodies, probes, or other molecules to enter nuclei while preserving their structural and genetic contents. The process typically uses chemical detergents, enzymatic treatment, or related conditions to create temporary or selective openings in the nuclear membranes, with fixation and treatment parameters adjusted to limit damage. In genetics, nuclei permeabilization supports access to chromatin and DNA for fluorescence staining, in situ hybridization, immunolabeling, and enzyme-based assays. By improving molecular access, it enables researchers to examine genome organization, gene regulation, DNA-associated proteins, and nuclear architecture in greater detail.
