12-well Plate Format

The 12-well plate format is a laboratory vessel that divides a single plate into 12 separate wells for organizing parallel biological experiments. Each well functions as an independent culture or reaction chamber, allowing researchers to apply distinct cells, microbes, treatments, or controls while maintaining consistent handling across the plate. In immunology and infection research, this format supports manageable comparisons of immune-cell responses, pathogen exposure, antimicrobial treatments, and inflammatory conditions. The uneven loss of medium from microtiter plates affects the reproducibility of uniform multicellular tumor spheroid formation. Improving culture conditions to reduce significant medium loss will improve the reproducibility of spheroid formation and the results of spheroid-based assays using the liquid-overlay technique.

This method quantifies microbial abundance using a 96-well plate format to plate colony forming units (CFUs) and is applied to the Drosophila microbiome in whole fly homogenate samples. CFUs are counted with an automated image analysis software provided here.
