Pilocarpine Stimulation

Pilocarpine stimulation is a pharmacological method that induces salivary secretion, enabling controlled collection of saliva for biological analysis. As a muscarinic acetylcholine receptor agonist, pilocarpine activates parasympathetic signaling in salivary glands, increasing fluid production and facilitating standardized sampling when spontaneous saliva is insufficient. In immunology and infection research, stimulated saliva can be examined for antibodies, cytokines, immune cells, microbial components, and pathogen-associated nucleic acids. This approach supports studies of mucosal immunity, host responses to infection, disease biomarkers, and vaccine effects while improving sample availability and consistency across experimental subjects.

This manuscript describes a method of inducing status epilepticus by systemic pilocarpine injection and of monitoring spontaneous recurrent seizures in live animals using a wireless telemetry video and electroencephalogram system. This protocol can be utilized for studying the pathophysiologic mechanisms of chronic epilepsy, epileptogenesis, and acute seizures.
