Amber Suppression Technology

Amber suppression technology is a genetic code expansion method that enables cells to produce proteins containing noncanonical amino acids at defined positions. It uses an engineered transfer RNA and matching aminoacyl-tRNA synthetase to recognize the amber stop codon, UAG, and insert a selected amino acid instead of terminating translation. In immunology and infection research, this approach supports precise protein labeling, controlled activation or inactivation, and analysis of pathogen or host proteins involved in immune recognition and disease. By providing site-specific chemical and structural control, amber suppression helps researchers investigate protein function, host-pathogen interactions, and potential therapeutic targets.

Source: Kalveram, B. et al. Using Click Chemistry to Measure the Effect of Viral Infection on Host-Cell RNA Synthesis. J. Vis. Exp. (2013)This video demonstrates the use of click chemistry and fluorescence microscopy to visualize nascent RNA transcription in mammalian cells. It highlights how viral infection suppresses host transcription by comparing fluorescent signals between infected and control cells.

Human In Vitro Suppression as Screening Tool for the Recognition of an Early State of Immune Imbalance

Tregs are potent suppressors of the immune system. There is a lack of unique surface markers to define them, hence, definitions of Tregs are primarily functional. Here we describe an optimized in vitro assay capable of identifying immune imbalance in subjects at risk to develop T1D.

Source: Kalveram, B., et al. Using Click Chemistry to Measure the Effect of Viral Infection on Host-Cell RNA Synthesis. J. Vis. Exp. (2013)This video demonstrates the use of click chemistry and flow cytometry to measure virus-induced suppression of host-cell transcription. Mammalian cells are infected with a virus that expresses a nuclear virulence protein disrupting the host transcription factor complex, while control cells remain transcriptionally active. Incorporation of 5-ethynyl uridine...

Total Factor Productivity (TFP) measures the efficiency with which inputs are transformed into outputs in production. It is the essence of economic growth, driven by technological advancement. Consider the agricultural sector, where production requires vast amounts of human labor and work animals. Today, modern farm machinery and agriculture technologies have revolutionized how we cultivate crops and produce food much more efficiently. TP = A*f(K, L) TFP is represented as a multiplier 'A' in...
