Blood Cell Preparation

Blood cell preparation is the set of laboratory methods used to isolate, clean, and preserve specific blood cell populations for analysis or experimentation. Procedures typically combine anticoagulation, centrifugation, density-gradient separation, selective red blood cell lysis, and washing to exploit differences in cell density, size, or membrane sensitivity while maintaining cell viability. Prepared whole blood, peripheral blood mononuclear cells, leukocytes, or erythrocytes support immunological assays, flow cytometry, microscopy, and infection studies. Consistent preparation improves measurements of immune-cell abundance and function, pathogen interactions, inflammatory responses, and treatment effects, making it essential for reproducible clinical and research workflows.

Establishment of human models of the blood-brain barrier (BBB) can benefit research into brain conditions associated with BBB failure. We describe here an improved technique for preparation of a contact BBB model, which permits coculturing of human astrocytes and brain endothelial cells on the opposite sides of a porous membrane.

Such preparations, when immunofluorescently stained with specific antibodies, enable us to study localization of proteins and identification of cell types within the entire vascular wall by confocal microscopy.

This protocol describes a detailed method for efficient generation of integration-free iPSCs from human adult peripheral blood cells. With the use of four oriP/EBNA-based episomal vectors to express the reprogramming factors, KLF4, MYC, BCL-XL, or OCT4 and SOX2, thousands of iPSC colonies can be obtained from 1 mL of peripheral blood.
