Bone Marrow Derived Cultured Mast Cells

Bone marrow-derived cultured mast cells (BMMCs) are laboratory-generated mast cells differentiated from hematopoietic progenitors in bone marrow, providing a renewable model for studying mast-cell biology. In culture, progenitor cells are expanded and guided toward the mast-cell lineage with growth factors such as interleukin-3, then assessed for markers including c-Kit and FcεRI; IgE-dependent antigen cross-linking can trigger degranulation and mediator release. BMMCs enable controlled analysis of histamine, cytokine, and lipid mediator responses in allergy, inflammation, and infection. They also support investigations of mast-cell roles in host defense and evaluation of immune-modulating compounds.

This video demonstrates the procedure for differentiating myeloid dendritic cells from mouse bone marrow. Isolation of mouse tibia and femur, and processing of bone marrow are demonstrated.
