Cellulose Paper Discs

Cellulose paper discs are small, absorbent pieces of purified cellulose used to collect, carry, or release defined amounts of biological and chemical samples in laboratory assays. In infection research, discs can be impregnated with antimicrobial agents or specimens and placed on a microbial culture; as the substance diffuses through agar, it may produce a measurable zone of growth inhibition that reflects antimicrobial activity. These discs support standardized disk-diffusion testing, screening of disinfectants and natural products, and evaluation of pathogen susceptibility. Their low cost, uniform size, and simple handling make them useful for teaching laboratories, diagnostic workflows, and comparative immunology and infection studies.

Covalent Binding of Antibodies to Cellulose Paper Discs and Their Applications in Naked-eye Colorimetric Immunoassays

In this manuscript, periodate oxidation and glutaraldehyde cross-linking methods for covalent immobilization of antibodies on paper discs are presented. The binding activity of immobilized antibodies was evaluated. Based on these results, a glutaraldehyde cross-linking method was used to develop a novel paper-based immunoassay for immunoglobulin G (IgG) detection.

The video demonstrates a paper-based immunoassay, involving amine-functionalized cellulose discs cross-linked with glutaraldehyde. Covalent immobilization of IgG-Fc-specific capture antibodies, followed by the introduction of immunoglobulin G and horseradish peroxidase-conjugated detection antibodies, results in a color change confirming the presence of immunoglobulin G.
