Colorimetric Reading

Colorimetric reading is a measurement approach that determines the concentration or presence of a target by analyzing the color produced in a chemical or biological assay. In immunology and infection research, an enzyme-linked reaction converts a colorless chromogenic substrate into a colored product, whose intensity is measured as absorbance with a spectrophotometer or plate reader. Comparing the signal with controls or a calibration curve allows researchers to estimate antibody, antigen, or microbial components in a sample. This method supports immunoassays, pathogen detection, and quantitative analysis because it provides a relatively simple and scalable way to translate molecular recognition into measurable data.

This video describes a colorimetric assay for lactate measurement in cell lysates by using the lactate dehydrogenase enzyme.
