Confocal Screening

Confocal screening is a fluorescence-based imaging approach that combines optical sectioning with systematic image acquisition to examine cells, tissues, or microorganisms in three dimensions. A focused laser scans fluorescently labeled specimens, while a pinhole rejects out-of-focus light and produces sharp optical slices that can be reconstructed into volumetric images. In immunology and infection research, this method reveals pathogen localization, host-pathogen interactions, immune-cell behavior, and changes in cellular structure. Quantitative analysis of captured images can measure infection burden, protein distribution, and treatment responses, supporting investigations of disease mechanisms and evaluation of antimicrobial or immunomodulatory interventions.

The complete construction of a custom, real-time confocal scanning imaging system is described. This system, which can be readily used for video-rate microscopy and microendoscopy, allows for an array of imaging geometries and applications not accessible using standard commercial confocal systems, at a fraction of the cost.
