Ex Vivo Generation

Ex vivo generation is the production, differentiation, or expansion of biological cells outside an organism under controlled laboratory conditions, providing a way to study immune responses without relying solely on whole-animal experiments. In immunology and infection research, investigators isolate primary cells or precursor populations and culture them with defined media, cytokines, growth factors, antigens, or other signals that regulate proliferation and cell fate, then evaluate their phenotype and function. These generated cell populations support studies of host-pathogen interactions, immune-cell development, vaccine responses, and therapeutic strategies, while enabling controlled comparison of cellular mechanisms and responses to infection.

Genes can be manipulated during development of the cortex or the hippocampus of the rat via in utero electroporation (IUE) at E16, to enable fast and targeted modifications in neuronal connectivity for later studies of behavior or neuropathology in adult animals. Postnatal in vivo imaging for control of IUE success is performed by bioluminescence of activating co-transfected luciferase.
