Fluorescent Bacteria

Fluorescent bacteria are bacterial cells that emit visible light after producing fluorescent molecules, providing a direct way to visualize microbial location, abundance, and behavior. In many research systems, bacteria express reporter genes such as those encoding green fluorescent protein; excitation at a specific wavelength causes the protein to emit light at a longer wavelength, allowing detection by fluorescence microscopy or flow cytometry. In immunology and infection research, fluorescent bacteria help track colonization, tissue invasion, phagocytosis, and immune-mediated clearance. These reporters connect microbial dynamics with host responses and support quantitative studies of infection, antimicrobial activity, and pathogen-host interactions.

Visualizing Bacteria in Nematodes using Fluorescent Microscopy

To study the mutualism between Xenorhabdus bacteria and Steinernema nematodes, methods were developed to monitor bacterial presence and location within nematodes. The experimental approach, which can be applied to other systems, entails engineering bacteria to express the green fluorescent protein and visualizing, using fluorescence microscopy bacteria within the transparent nematode.
