Heme Peroxidase Inhibition

Heme peroxidase inhibition is the reduction or prevention of enzymatic reactions carried out by heme-containing peroxidases, which use hydrogen peroxide to oxidize target molecules. Inhibition can occur when a compound binds the enzyme’s heme center, competes with substrates, alters peroxide-dependent catalytic steps, or disrupts formation of reactive intermediates. In immunology and infection research, studying this process helps clarify how peroxidases contribute to antimicrobial defense, inflammatory signaling, and tissue injury. Inhibitor assays also support the characterization of host and microbial enzymes, identification of regulatory compounds, and evaluation of how pathogens may evade or withstand oxidative immune mechanisms.

Altered intracellular heme levels are associated with common diseases such as cancer. Thus, there is a need to measure heme biosynthesis levels in diverse cells. The goal of this protocol is to provide a fast and sensitive method to measure and compare the levels of heme synthesis in different cells.

This protocol describes the quick enrichment of leukocytes from small blood samples for a subsequent specific determination of the halogenating peroxidase activity within the cells. The method can be applied to human and non-human material and may contribute to the evaluation of new inflammatory markers.

Bathophenanthroline Sulfonate-Based Colorimetric Assay: A Simple and Rapid Method for Quantitation of Non-Heme Iron in Mouse Liver Tissue

In this video, we describe a colorimetric-based method to determine non-heme iron content in mouse liver tissue. Bathophenanthroline sulfonate is used as the chromogenic substrate in the presence of a reducing agent, which converts ferric iron to chelatable ferrous iron.

Gua Sha, traditional Chinese therapeutic skin scraping, causes subcutaneous microvascular blood extravasation. We report a protocol of bioluminescence imaging of HO-1-luciferase transgenic mice to demonstrate that Gua Sha upregulates heme oxygenase-1 (HO-1) in multiple organs.

This video demonstrates an assay measuring influenza neuraminidase (NA) inhibiting antibody titers in a serum sample. A serum sample containing antibodies against NA is added to a fetuin-coated microwell plate.
