Human Serum Dilution

Human serum dilution is a laboratory method that reduces the concentration of antibodies, complement proteins, and other serum components in a controlled, stepwise manner to support immunological testing. In a typical serial dilution, serum is mixed with a defined volume of diluent, often in successive twofold or tenfold steps, and each preparation is tested against a standardized antigen, pathogen-derived material, or assay reagent. The highest dilution that produces a defined response can indicate an antibody titer or relative immune reactivity, while dilution also helps reduce matrix effects and bring signals into an assay’s measurable range. In infection research, this approach supports serology, neutralization studies, complement-dependent assays, and comparison of immune responses across samples.

We have used this method to isolate microRNAs from cancer serum for use in DNA arrays and also singleplex quantitative PCR.
