Immunomagnetic Separation

Immunomagnetic separation is a technique that isolates specific cells, microorganisms, or other targets from complex samples using antibody-coated magnetic particles. The antibodies recognize surface markers on the target, allowing magnetic beads to bind selectively; applying a magnetic field then retains the labeled targets while unbound material is removed through washing. In immunology and infection research, this method supports the enrichment or depletion of immune cell populations, recovery of pathogens, and preparation of samples for flow cytometry, molecular assays, or culture. Its specificity and efficient handling of heterogeneous samples make it valuable for studying host responses and detecting infectious agents.

We present the techniques required to isolate the stromal vascular fraction (SVF) from mouse inguinal (subcutaneous) and perigonadal (visceral) adipose tissue depots to assess their gene expression and collagenolytic activity. This method includes the enrichment of Sca1high adipose-derived stem cells (ASCs) using immunomagnetic cell separation.
