In Vivo Antibody

In vivo antibodies are immunoglobulins produced within or administered to a living organism, where they recognize specific antigens and shape immune defense. Their antigen-binding regions attach to microbial or cellular targets, while Fc regions recruit mechanisms such as neutralization, opsonization, complement activation, and antibody-dependent cellular cytotoxicity. Studying antibody activity in vivo reveals how antibody concentration, tissue distribution, stability, and interactions with immune cells influence protection against infection. These principles support passive immunization, therapeutic antibody development, vaccine evaluation, and the design of treatments that improve pathogen clearance while limiting harmful immune responses.

A Technique to Assess the In Vivo Localization of Tumor-Specific Antibodies

2025

This video demonstrates a technique to study the in vivo localization of tumor-specific antibodies in a mouse tumor xenograft model. Upon subcutaneously injecting cancer cells into mice for tumor formation, fluorescently-labeled antibodies are injected that bind to tumor-specific receptors, enabling in vivo assessment of antibody localization via imaging.

An In Vivo Technique to Track Bispecific Antibody-Induced T Cell Trafficking to Tumors

2025

This video demonstrates a method for tracking bispecific antibody-induced T cell trafficking to tumors in a mouse model. Luciferase-transduced effector T cells are injected into a mouse with a subcutaneous tumor cell xenograft. Bispecific antibodies are then introduced, binding to T cells and the tumor cells. The localization of the T cells is tracked using bioluminescent imaging, with the magnitude of the signal indicating the extent of T cell infiltration into the tumor.

In Vivo Fluorescence Imaging to Localize Antibodies in a Mouse Tumor Xenograft Model

2025

The video demonstrates a fluorescence imaging method for in vivo antibody localization in a mouse tumor xenograft model. It involves the injection of tumor cells in a mouse to generate solid xenografts beneath the skin. Subsequent injection of fluorescent antibodies confirms specific localization through enhanced fluorescence signals within the tumor xenograft.

2025

This video demonstrates an in vivo immunofluorescence localization (IVIL) technique to study the biodistribution of tumor-targeting antibodies in a murine model of breast cancer. Upon the intravenous injection of a tumor-specific antibody in the mouse, the tumor is surgically isolated, and the localization of the antibody within the tumor tissue is confirmed via ex vivo immunostaining.
