Influenza A Virus Infection

Influenza A virus infection is a respiratory illness caused by an enveloped, segmented RNA virus that infects airway epithelial cells and stimulates innate and adaptive immune responses. Viral hemagglutinin binds sialic acid receptors to promote cell entry, while endosomal acidification triggers membrane fusion and release of the viral genome; the virus then replicates its RNA and assembles new particles for release. Studying this process in immunology and infection clarifies how interferons, antibodies, T cells, and inflammatory responses control disease, while also informing vaccine design, antiviral development, and surveillance of emerging strains.

Influenza A viruses (IAVs) are important human respiratory pathogens. To understand the pathogenicity of IAVs and to perform preclinical testing of novel vaccine approaches, animal models mimicking human physiology are required. Here, we describe techniques to evaluate IAV pathogenesis, humoral responses and vaccine efficacy using a mouse model of infection.

Purified viral complexes can be analyzed by both Western blot and primer extension of protein and RNA content, respectively.
