Intracellular Amastigotes

Intracellular amastigotes are the tissue-resident, replicative form of Leishmania parasites that develops within host cells, especially macrophages, where it drives disease and challenges immune defense. After transmission by an infected sand fly, extracellular promastigotes are engulfed and transform into amastigotes adapted to the acidic, enzyme-rich phagolysosomal environment; they multiply by binary fission while modulating host-cell signaling and antimicrobial responses. Studying this stage clarifies parasite persistence, host-pathogen interactions, and mechanisms of immune evasion. Intracellular amastigote models also support diagnosis, evaluation of antiparasitic compounds, and research into vaccines and therapies for leishmaniasis.

A parasite-rescue and transformation assay with THP1 cells infected in vitro with Leishmania donovani has been optimized for anti-leishmanial drug screening. The assay involves differentiation of THP1 cells, infection with promastigotes, treatment with test drugs, controlled lysis of the infected macrophages, rescue of amastigotes, transformation to promastigotes and monitoring promastigote growth and proliferation with a fluorometric assay.

Isolation of Cell-Surface and Intracellular Proteins from an Astrocyte Culture

This video demonstrates the isolation of cell-surface and intracellular proteins from an astrocyte culture. The astrocyte culture is placed on ice to inhibit endocytosis, and then the cell-surface proteins are labeled with a biotinylation reagent. The cells are lysed to release the biotinylated cell-surface proteins and non-biotinylated intracellular proteins. Finally, streptavidin-coated beads are used to separate the biotinylated cell-surface proteins from the intracellular proteins.
