Latent Infection Establishment

Latent infection establishment is the process by which a pathogen enters and maintains a persistent, low-activity state in a host without causing continuous productive disease. This process depends on survival within protected cellular or tissue reservoirs, limited pathogen replication, and modulation of host immune detection. By balancing persistence against immune clearance, latent pathogens can remain clinically silent yet retain the capacity to reactivate when immune control weakens or host conditions change. Studying latent infection establishment helps explain chronic disease, transmission, treatment failure, and recurrence while guiding vaccine design, antiviral strategies, and approaches that target persistent reservoirs.

Establishment and Quantification of De Novo Lytic Infection by Cell-free Kaposi's Sarcoma-Associated Herpesvirus

2025

This protocol describes an in vitro system to model de novo lytic infection of Kaposi's sarcoma-associated herpesvirus using BAC16-derived virions. The method enables investigation of early viral replication and dissemination. Infectious virus production is quantified by GFP-based infectious units assays and real-time PCR of encapsidated viral genomes.

Establishing a Mouse Model of Vaginal Group B Streptococcus Infection

Source: Patras, K. A., Doran, K. S. A Murine Model of Group B Streptococcus Vaginal Colonization. J. Vis. Exp. (2016)This video demonstrates the preparation and vaginal inoculation of a hormone-treated female mouse with Group B Streptococcus to model infection. It explains how to culture and prepare the bacteria, administer the inoculum into the vaginal lumen, and ensure successful infection.

Establishing an Active HIV Infection in a Rat Model

2025

This video demonstrates the procedure of injecting chimeric HIV viruses into the cortex of a rat's brain, where the virus RNA integrates into the host genome, replicates, and spreads to establish an active HIV infection in the experimental model.

Establishing Bacterial Infection in Drosophila Larvae

Source: Hiroyasu, A., D et. al., Extraction of Hemocytes from Drosophila melanogaster Larvae for Microbial Infection and Analysis. J. Vis. Exp. (2018)This video demonstrates the procedure for establishing a bacterial infection in Drosophila larvae by placing them into a bacterial droplet and pricking them with a tungsten needle. The larvae are then incubated on a grooved, nutrient-rich agar plate to study host-pathogen interactions.

Establishment of a Urinary Tract Infection Model in Female Mice

2025

This video demonstrates the establishment of uropathogenic Escherichia coli infection in a female mouse bladder through transurethral catheter instillation. The injected bacteria successfully establish a urinary tract infection within the bladder, providing a valuable platform for studying healthcare-associated diseases related to the urinary tract.
