Clinical Isolates

Clinical isolates are microorganisms recovered from patient specimens and maintained for laboratory study, providing direct evidence of the pathogens associated with human infection. Laboratories obtain them from samples such as blood, respiratory secretions, or tissue, then use selective culture conditions and microbiological or molecular methods to isolate and identify individual strains. In immunology and infection research, clinical isolates support analysis of host-pathogen interactions, immune recognition, virulence, and antimicrobial susceptibility. Because they retain genetic and phenotypic diversity found in patients, these isolates help evaluate diagnostic methods, compare immune responses, and improve the clinical relevance of infection models.

Here we present a novel clinical grade isolation and culture method for kidney Perivascular Stromal Cells (kPSCs) based on whole organ perfusion with digestive enzymes and NG2-cell enrichment. With this method, it is possible to acquire sufficient cell numbers for cellular therapy.

A protocol is described here that enables clinical support staff to effectively process and store cutaneous tumor (e.g., squamous cell carcinoma, basal cell carcinoma, and melanoma) samples for downstream laboratory applications without interfering with clinical operations.

Isolation of Brain-infiltrating Leukocytes

A rapid method to obtain infiltrating leukocytes from the murine brain is described. This method utilizes a continuous Percoll gradient and discontinuous Ficoll gradient to select and purify the leukocyte-enriched layer. Isolated leukocytes may then be characterized by flow cytometric measurements.
