Mouse Holder Preparation

Mouse holder preparation is the process of readying a restraint device and positioning a mouse for controlled, consistent handling during experimental procedures. The holder limits body movement while keeping the animal appropriately aligned and providing access to the intended site, which helps reduce procedural variation and supports safe manipulation. In immunology and infection research, prepared holders can facilitate inoculation, blood collection, tissue sampling, and other interventions that require stable positioning. A procedure for making en face preparations of the mouse carotid artery and aorta is described. Such preparations, when immunofluorescently stained with specific antibodies, enable us to study localization of proteins and identification of cell types within the entire vascular wall by confocal microscopy.

We describe a protocol for transcardiac perfusion of mice, removal and sectioning of the brain, as well as immunoperoxidase staining, resin embedding, and ultrathin sectioning of the brain sections. Upon completion of these procedures, the immunostained material is ready for examination with transmission electron microscopy.
