Mouse Paw Processing

Mouse paw processing is a tissue-preparation method used to examine localized inflammation, infection, and immune responses in the extremities of laboratory mice. The procedure typically involves collecting the paw tissue, removing unwanted material, and mechanically disrupting or homogenizing the sample so that cells, proteins, nucleic acids, or infectious agents can be recovered for analysis. Processed tissue supports measurements of immune-cell infiltration, pathogen burden, cytokine production, and inflammatory pathology using techniques such as flow cytometry, culture, microscopy, or molecular assays. Standardized handling and timing help preserve biological signals and enable consistent comparisons between experimental groups.

Classical forelimb asymmetry analysis of the cylinder test is routinely used to assess behavioural deficits in rats following brain injury or stroke; however, it fails to detect consistent deficits in mice. This study demonstrates that quantifying paw-dragging behaviour is a more sensitive analysis of brain injury in mice.
