Peritoneal Leukocytes

Peritoneal leukocytes are immune cells located within the abdominal peritoneal cavity, where they provide local surveillance and coordinate responses to infection, injury, and inflammation. Resident macrophages and lymphocytes monitor the cavity, while signals such as chemokines and inflammatory cytokines can recruit circulating monocytes and neutrophils across the mesothelial barrier to eliminate microbes and remove damaged material. Characterizing these cell populations helps researchers understand host defense, immune regulation, and inflammatory diseases affecting the abdomen. Their analysis also supports studies of infection models, peritoneal inflammation, and therapies that alter leukocyte recruitment or function.

Marginating-pulmonary leukocytes exhibit unique characteristics and distinct immunological functions compared to other leukocyte populations. Here we describe selective harvesting of this subpopulation of pulmonary leukocytes, by forced perfusion of the lungs in rats and mice. Marginating-pulmonary leukocytes seem critical in determining susceptibility to blood-borne and lung-related diseases.

Described herein is a protocol to isolate and analyze the infiltrating leukocytes of tissues at the maternal-fetal interface (uterus, decidua, and placenta) of mice. This protocol maintains the integrity of most cell surface markers and yields enough viable cells for downstream applications including flow cytometry analysis.

Isolation of Brain-infiltrating Leukocytes

A rapid method to obtain infiltrating leukocytes from the murine brain is described. This method utilizes a continuous Percoll gradient and discontinuous Ficoll gradient to select and purify the leukocyte-enriched layer. Isolated leukocytes may then be characterized by flow cytometric measurements.
