Platelet Activating Factor

Platelet-activating factor (PAF) is a potent phospholipid mediator that coordinates inflammation, vascular responses, and immune cell activity during infection and tissue injury. Cells synthesize PAF on demand, and it binds a specific G protein-coupled receptor on platelets, leukocytes, and vascular cells to trigger platelet aggregation, leukocyte activation, chemotaxis, and increased vascular permeability. In immunology and infection research, PAF helps explain how inflammatory signals recruit immune cells and alter blood vessel function, while excessive PAF signaling can contribute to tissue damage and systemic inflammation. Studying this pathway supports research into host-pathogen interactions and potential anti-inflammatory interventions.

Purification and Activation of Platelets from Murine Whole Blood

2025

In this video, we describe a protocol for purifying platelets from murine whole blood and their subsequent activation with the platelet agonist thrombin. When analyzed with flow cytometry, the purified resting platelets are characterized based on the expression of their cell-surface marker CD41, whereas purified activated platelets co-express both the surface markers CD41 and CD62P.

This article outlines the protocol for quantifying the percentage of Tissue Factor (TF)-positive platelets using whole blood flow cytometry, assessing the protein: (1) intracellularly in resting conditions, and (2) on the cell surface, in both resting and activated conditions. Guidance is also provided for evaluating TF-positive platelets in platelet-rich plasma.

We describe a straightforward method for the isolation of washed platelets from human blood followed by agonist-induced platelet aggregation measurements by turbidimetry. As an example we apply this method for studying the aggregation response of human platelets to collagen after a pre-incubation with the Pannexin1 channel inhibitor Brilliant Blue FCF.

A Microfluidic Flow Chamber Model for Platelet Transfusion and Hemostasis Measures Platelet Deposition and Fibrin Formation in Real-time

This paper describes an experimental model of hemostasis that simultaneously measures platelet function and coagulation. Platelet and fibrin fluorescence is measured in real-time, and platelet adhesion rate, coagulation rate, and onset of coagulation are determined. The model is used to determine platelet procoagulant properties under flow in concentrates for transfusion.
