Recombinant Antibody Production

Recombinant antibody production is the process of generating engineered immunoglobulins from cloned antibody genes, providing a consistent alternative to antibodies obtained directly from immune cells. Researchers isolate or design sequences encoding antibody heavy and light chains, insert them into expression vectors, and introduce these constructs into host cells that assemble and secrete the antibody for purification. In immunology and infection research, recombinant antibodies support precise pathogen detection, antigen characterization, diagnostic assay development, and therapeutic design. Their defined sequences enable reproducible manufacturing, controlled affinity or specificity changes, and rapid adaptation to emerging infectious agents.

An efficient system of structure and function analysis of a gene in an ex vivo culture of splenic B-lymphocytes is described. This method takes advantage of recombinant retroviral production in a helper free, ecotrophic packaging cell line. Stable, heritable expression of a gene of interest within primary lymphocytes is achieved leading to generation of surface antibodies on B cells undergoing class switch recombination.

A method is described with visual accompaniment for conducting scalable, high throughput selections from phage-displayed combinatorial synthetic antibody libraries against hundreds of antigens simultaneously. Using this parallel approach, we have isolated antibody fragments that exhibit high affinity and specificity for diverse antigens that are functional in standard immunoassays.
