Resazurin Viability Assay

The resazurin viability assay is a cell-based method for estimating the number of living, metabolically active cells, making it useful for assessing cellular health and survival. In the assay, viable cells reduce the blue, nonfluorescent indicator resazurin to pink, fluorescent resorufin through intracellular metabolic activity, producing a signal that can be measured by fluorescence or absorbance. In immunology and infection research, this approach supports evaluation of immune-cell viability, microbial growth, and the effects of infectious agents or candidate treatments. Its simple readout enables comparisons across experimental conditions and helps quantify changes in cellular activity without directly counting individual cells.

Most bacterial infections produce a biofilm. By virtue of their environment, biofilm associated bacteria are often phenotypically drug resistant. Novel antibacterial molecules that kill bacteria in biofilms are thus a high priority. We establish an assay to quickly screen for antimicrobial compounds that are effective at eradicating biofilms.

Therapeutic compounds are often first examined in vitro with viability assays. Blind cell counts by a human observer can be highly sensitive to small changes in cell number but do not assess function. Computerized viability assays, as described here, can assess both structure and function in an objective manner.
