Tnf-alpha Stimulation

TNF-alpha stimulation is the experimental activation of cells with tumor necrosis factor alpha, a proinflammatory cytokine that regulates immune defense, inflammation, and cell survival. When TNF-alpha binds TNF receptors, particularly TNFR1 or TNFR2, it triggers intracellular signaling through adaptor proteins and pathways such as NF-kB and MAPK, altering gene expression and cellular behavior. In immunology and infection research, this approach helps model inflammatory responses, examine host-pathogen interactions, and assess how immune cells respond to microbial signals or therapeutic interventions. It can also clarify mechanisms underlying tissue damage, chronic inflammation, and immune-mediated disease.

Notch signaling is a form of cellular communication that relies upon direct contact between cells. To properly induce Notch signaling in vitro, Notch ligands must be presented to cells in an immobilized state. This protocol describes methods for in vitro stimulation of Notch signaling in mouse osteoclast precursors.
