Uveitis Mouse Model

An uveitis mouse model is an in vivo research system used to reproduce inflammation of the uvea, the eye’s vascular middle layer, and to study how immune responses damage ocular tissues. Depending on the model, researchers trigger disease through autoimmune recognition of retinal or uveal antigens, infection, or other inflammatory stimuli, then monitor leukocyte infiltration, blood-retinal barrier disruption, cytokine signaling, and tissue injury. These models help clarify mechanisms of ocular inflammation, evaluate immunomodulatory or antimicrobial treatments, and identify biomarkers of disease activity. Their controlled genetics, timing, and tissue sampling also support comparisons between innate and adaptive immune pathways relevant to human uveitis.

In this report we present a protocol that allows the investigator to generate a mouse model of intraocular uveitis. More commonly referred to as experimental autoimmune uveitis (EAU), this established model captures many aspects of human disease. Herein, we will describe how to induce and monitor disease progression using several readouts.

This protocol outlines the steps for inducing Primed Mycobacterial Uveitis (PMU) in mice. This method outlines the steps to help produce reliable and robust ocular inflammation in the mouse model system. Using this protocol, we generated uveitic eyes and uninflamed fellow eyes from single animals for further evaluation with immunologic, transcriptomic, and proteomic assays.

Mouse Models of Periventricular Leukomalacia

We established mouse models of periventricular leukomalacia (PVL), the predominant brain injury in premature infants characterized by periventricular white matter lesions. Hypoxia/ischemia with/without systemic infection are the primary causes of PVL. Unilateral carotid ligation and hypoxia exposure with/without lipopolysaccharide injection creates PVL-like lesions in P6 mice.
