Adrenal Sarcoma

Adrenal sarcoma is a rare malignant tumor arising from mesenchymal, or connective-tissue, cells in the adrenal gland or surrounding adrenal region, and it matters because its rarity and aggressive behavior can complicate diagnosis and treatment. These tumors develop when abnormal cells proliferate unchecked, form a mass, invade nearby structures, and may spread through blood or lymphatic pathways; imaging identifies the lesion, while histopathology and immunohistochemistry help determine its type and origin. Management depends on tumor subtype, extent, and resectability, with surgery often central when complete removal is feasible and systemic therapy or radiation considered in selected cases. Studying adrenal sarcoma supports more accurate classification and individualized care.

The In ovo CAM-assay as a Xenograft Model for Sarcoma

The in ovo chorioallantoic membrane (CAM) is grafted with fresh sarcoma-derived tumor tissues, their single cell suspensions, and permanent and transient fluorescently labeled established sarcoma cell lines. The model is used to study graft- (viability, Ki67 proliferation index, necrosis, infiltration) and host (fibroblast infiltration, vascular ingrowth) behavior.

We present here a cell culture method for inducing mesenchymal-epithelial transitions (MET) in sarcoma cells based on combined ectopic expression of microRNA-200 family members and grainyhead-like 2 (GRHL2). This method is suitable for better understanding the biological impact of phenotypic plasticity on cancer aggressiveness and treatments.

A Novel Method: Super-selective Adrenal Venous Sampling

'Super-selective' adrenal venous sampling (ssAVS, also called segmental adrenal venous sampling: sAVS) was performed using a micro-catheter to identify adrenal segment(s) that produce excessive amounts of hormones. The ssAVS technique was described, cases in which adrenal segmental lesion(s) were identified by ssAVS were presented, and the usefulness of ssAVS in future adrenal research was discussed.

Host cell factors play a critical role in the establishment and maintenance of Kaposi's sarcoma (KS). We outline methods to identify host cell factors altered in KSHV-infected DMVEC cells, and in KS tumor tissue. Cellular genes altered by virus will serve as potential target(s) for novel therapeutics.

Adrenal activity can be assessed in the equine species by analysis of feces for corticosterone metabolites. The method offers a non-invasive option to assess long term patterns in both domestic and free ranging horses. This protocol describes the enzyme linked immunoassay involved and the associated biochemical validation.
