Ascending Aortic Banding

Ascending aortic banding is a surgical technique that creates controlled pressure overload on the heart by narrowing the ascending aorta, making it an important model for studying cardiac adaptation and disease. A constrictive band increases left ventricular afterload, requiring the ventricle to generate greater pressure to eject blood and often triggering compensatory hypertrophy, followed by ventricular remodeling or failure if the overload persists. In medicine and cardiovascular research, this approach helps investigators examine the progression of pressure-overload heart disease, identify molecular and functional changes, and evaluate potential treatments for hypertrophy, remodeling, and heart failure.

Ascending Aortic Constriction in Rats for Creation of Pressure Overload Cardiac Hypertrophy Model

We describe a stepwise procedure for creating pressure overload and left ventricular hypertrophy in Wistar rats by constriction of the ascending aorta using a small metallic clip. This model is extensively used for studying remodeling changes during cardiac hypertrophy and for identifying and evaluating strategies for regression of such changes.

Measuring Ascending Aortic Stiffness In Vivo in Mice Using Ultrasound

We describe a technique for measuring aortic stiffness from its pressure-diameter relationship in vivo in mice. Aortic diameter is recorded by ultrasound and aortic pressure is measured invasively with a solid-state pressure catheter. Blood pressure is changed incrementally and the resulting diameter is measured.

The present protocol describes a new technique of aortic banding in mice to induce pressure overload cardiac hypertrophy. For banding, a rubber ring with a fixed inner diameter is used. This new technique promises less variance and more reproducible data for future experiments.
