Athymic Nude Mouse

An athymic nude mouse is a hairless, immunodeficient laboratory mouse that lacks a functional thymus, making it an important model in medical research. A mutation affecting the Foxn1 gene disrupts thymic development and greatly reduces the production of mature T lymphocytes, allowing transplanted human or animal cells to survive with limited immune rejection. Researchers use these mice to establish tumor xenografts, evaluate cancer therapies, study transplantation, and investigate interactions between grafted tissues and the host environment. Their predictable immune deficiency supports controlled experiments, although residual immune functions and differences between mice and humans must be considered when interpreting results.

Mycobacterium leprae, the causative agent of leprosy, does not grow in vitro. We describe an easy to follow protocol to prepare a bacillary suspension to ensure the maintenance of large quantities of M. leprae for a variety of applications. Protocols for propagation by mouse footpad inoculation, evaluation of viability, freezing and thawing bacillary stock are described in detail.
