Cerebral Region Collection

Cerebral region collection is the systematic isolation of defined anatomical areas of the brain for examination, comparison, or further laboratory analysis. The process uses anatomical landmarks and precise dissection to separate regions while preserving their tissue architecture, cellular composition, and molecular contents; collected samples may then undergo imaging, histology, biochemical testing, or molecular profiling. In medicine, this approach helps researchers relate regional structure to neural function, disease-associated changes, and treatment responses. Consistent collection methods also improve comparisons among specimens, support studies of brain development and pathology, and provide material for investigating region-specific biomarkers and mechanisms.

Protein synthesis is a critical biological process for cells. In brain, it is required for adaptive changes. Measurement of rates of protein synthesis in the intact brain requires careful methodological considerations. Here we present the L-[1-14C]-leucine quantitative autoradiographic method for determination of regional rates of cerebral protein synthesis in vivo.

Construction and Application of Cerebral Functional Region-Based Cerebral Blood Flow Atlas Using Magnetic Resonance Imaging-Arterial Spin Labeling

2024

This study integrated magnetic resonance imaging- arterial spin labeling images to derive cerebral blood flow (CBF) atlas for cerebral functional regions. Comparing typical healthy and chronic cerebral ischemia CBF atlases revealed significant differences in regional CBF distributions, enabling rapid, noninvasive assessments of functional CBF to assist in diagnosis and evaluate therapeutics.

Surgical induction of ischemic brain damage in the rat is a widely used model for stroke research. Here we demonstrate the induction of focal cerebral ischemia by occlusion of the middle cerebral artery. Visualization of the resulting infarct by histological staining and magnetic resonance imaging is also shown.

Use of Cerebral Open-Flow Microperfusion for the Longitudinal Collection of Interstitial Fluid in an Animal Model of Glioblastoma

2025

In vivo, intercellular communications underpinning many central nervous system diseases, such as glioblastoma (GBM), are notoriously difficult to measure and characterize. Here, we describe procedures of cerebral open-flow microperfusion (cOFM) that can be employed to sample interstitial fluid components in a longitudinal animal model of GBM.
