Corneal Wound Healing

Corneal wound healing is the coordinated biological process by which the eye repairs damage to the corneal epithelium, stroma, or endothelium, preserving transparency and vision. After injury, epithelial cells spread and migrate across exposed surfaces, proliferate to restore coverage, while inflammatory signals recruit immune cells and activate stromal keratocytes; extracellular matrix remodeling and endothelial responses help re-establish tissue structure and fluid balance. Understanding this process supports treatment of abrasions, ulcers, and surgical injuries, while guiding evaluation of contact lenses and therapies designed to limit scarring, haze, and visual impairment.

Herein, we present a protocol to use heat shock protein 27 (HSP27)-specific small interfering RNA to assess the function of HSP27 during corneal epithelial wound healing. RNA interference is the best method for effectively knocking-down gene expression to investigate protein function in various cell types.

Here, a protocol for creating a central corneal epithelial abrasion wound in the mouse using a trephine and a blunt golf club spud is described. This corneal wound healing model is highly reproducible and is now being used to evaluate compromised corneal wound healing in the context of diseases.

This protocol focuses on damaging the ocular surface of zebrafish through abrasion to assess the subsequent wound closure at the cellular level.
