Endothelial Cell Model

An endothelial cell model is an in vitro system that represents the cells lining blood vessels, enabling controlled study of vascular structure and function. Cultured endothelial cells form cell-cell junctions and selectively regulate permeability, while responding to fluid shear stress, inflammatory mediators, and angiogenic signals. In medicine, these models help investigate vascular inflammation, thrombosis, tumor angiogenesis, and blood-brain barrier function, and support evaluation of drugs or biomaterials that affect vascular health. Depending on the design, models may use monolayers, three-dimensional cultures, or tissue-engineered vessels to improve physiological relevance and connect cellular mechanisms with disease outcomes.

Adaptive immunity is controlled by dynamic 'immunological synapses' formed between T cells and antigen presenting cells. This protocol describes methods for investigating endothelial cells both as understudied physiologic APCs and as a novel type of 'planar cellular APC model'.

We synthesized and characterized a tunable gelatin-based substrate for culturing vascular endothelial cells (ECs) under relevant vascular flow conditions. This biomimetic surface replicates both physiological and pathological conditions, enabling the study of mechanical forces on EC behavior and advancing our understanding of vascular health and disease mechanisms.
