Ex Vivo Tissue Model

An ex vivo tissue model is a laboratory system that preserves a tissue removed from an organism so researchers can study its structure and function under controlled conditions. The tissue is maintained in culture media with appropriate nutrients, oxygen, temperature, and signaling factors, allowing key cellular interactions and physiological responses to continue outside the body. In medicine, ex vivo tissue models help investigate disease mechanisms, evaluate drug or device effects, and assess tissue injury and repair. Because they retain aspects of native architecture and cell communication, these models can provide biologically relevant evidence while reducing reliance on whole-animal studies and supporting the development of personalized treatments.

Here, we present a systematic approach for developing physiologically relevant, sensitive and specific in vivo assays for interpreting variation in human pathology. Transient genetic manipulation via microinjection of WT and mutant human mRNA and morpholino (MO) antisense oligonucleotides harness the tractability of the developing zebrafish embryo to rapidly assay pathogenic mutations, especially, but not exclusively, in the context of human developmental disorders.
