Expansion Pathology

Expansion pathology refers to disease caused by abnormal enlargement of repetitive DNA sequences, a genetic mechanism underlying several inherited disorders. During DNA replication or repair, unstable tandem repeats can expand beyond a disease-associated threshold, disrupting gene expression, altering RNA processing, or producing toxic proteins; repeat length may also increase across generations, a phenomenon known as genetic anticipation. In medicine, recognizing expansion pathology supports molecular diagnosis, risk assessment, and interpretation of variable age of onset and disease severity. Testing methods such as repeat-primed PCR and long-read sequencing can identify expansions that standard sequencing may miss, improving evaluation of neurological, muscular, and multisystem genetic disease.
