Histological Parenchymal Biopsies

Histological parenchymal biopsies are tissue samples taken from an organ’s functional tissue and examined microscopically to identify structural and cellular abnormalities. A clinician collects a small specimen, which is preserved, processed into thin sections, stained, and evaluated for changes in tissue architecture, cell morphology, inflammation, fibrosis, or other disease-associated features. In medicine, these biopsies support diagnosis, classification, and assessment of conditions affecting organs such as the liver, kidney, lung, and gastrointestinal tract. Findings can help distinguish disease processes, estimate severity, guide treatment decisions, and provide tissue-based evidence for research into disease mechanisms and therapeutic response.

This manuscript describes a simple and reproducible protocol for isolation of intracerebral arterioles (a group of blood vessels encompassing parenchymal arterioles, penetrating arterioles and pre-capillary arterioles) from mice, to be used in pressure myography, immunofluorescence, biochemistry, and molecular studies.

This video demonstrates a method for isolating the endothelial tubes from mouse cerebral parenchyma. Through a combination of enzymatic digestion and mechanical dissociation, the process yields an intact tube with endothelial cells.

Tools used for visualizing vascular regeneration require methods for contrasting the vascular trees. This film demonstrated a delicate injection technique used to achieve optimal contrasting of the vascular trees and illustrate the potential benefits resulting from a detailed analysis of the resulting specimen using µCT and histological serial sections.

Here, we present a standardized protocol for detecting prostate cancer using stimulated Raman histology (SRH) on biopsy samples due to advantages over traditional histopathology, focusing on sample preparation, imaging, and artificial intelligence to improve the cancer-to-tissue ratio. Additionally, it supports biobanking for transcriptomics, organoid or xenograft models, and identification of intraoperative surgical margins.

We standardized an abdominal adipose tissue biopsy using a non-diathermy method performed under local anesthesia. Three cases of excessive post-operation bleeding out of 115 operations (2.61%) occurred. We conclude that an abdominal adipose tissue surgical biopsy using a non-diathermy method can be safely applied to healthy men.
