Laser-assisted Phosphorimetry

Laser-assisted phosphorimetry is an analytical technique that uses laser light to excite phosphorescent molecules and measure their delayed light emission, providing quantitative information about chemical or biological environments. In medical research, a laser pulse activates an oxygen-sensitive phosphor, and the emitted phosphorescence is recorded after excitation; oxygen-dependent quenching changes the emission lifetime, allowing calculation of local oxygen concentration or tissue oxygen tension. This approach supports noninvasive or minimally invasive assessment of oxygenation in tissues, blood, and cell systems. Its measurements can help characterize hypoxia, evaluate circulation and metabolism, and improve studies of disease mechanisms, treatment response, and therapeutic technologies.

Here we present a protocol for laser-assisted microdissection of specific plant cell types for transcriptional profiling. While the protocol is suitable for different species and cell types, the focus is on highly inaccessible cells of the female germline important for sexual and apomictic reproduction in the crucifer genus Boechera.
