Rabbit Hindlimb Model

The rabbit hindlimb model is an in vivo research system that uses the rabbit leg to investigate musculoskeletal, vascular, and tissue-repair processes relevant to medicine. Researchers apply a controlled intervention, such as injury, altered blood flow, surgical reconstruction, or mechanical loading, and then assess changes in tissue structure and function over time. Because the model supports anatomical, histological, imaging, and functional evaluation, it helps researchers examine disease mechanisms, healing responses, biomaterials, and potential therapies before clinical translation. Findings from this model can guide the development of treatments for limb injury, impaired circulation, bone and muscle disorders, and regenerative medicine.

Surgical induction of hindlimb ischemia in the mouse is useful to examine angiogenesis, however this is compromised in certain inbred mouse strains that display marked ischemia-induced tissue necrosis. Methods are described to induce subacute limb ischemia using ameroid constrictors to circumvent this problem through the induction of gradual arterial occlusion.

Anesthesia and Monitoring in a Rabbit Model

All animal procedures described here must be conducted in accordance with institutional animal ethics guidelines and approved by IACUC. All procedures must follow the principles of the 3Rs—Replacement, Reduction, and Refinement—and must be performed by trained personnel. Anesthesia is an essential component of rabbit surgical and experimental procedures because it prevents pain, reduces stress responses, and helps immobilize the animal during handling and manipulation when general anesthesia is...
