Shrna Screening

shRNA screening is a gene-silencing approach that systematically identifies how reduced expression of selected genes affects a cellular or disease-related phenotype. It uses short hairpin RNA sequences, typically delivered by vectors, that are processed into small interfering RNA and guide the RNA-induced silencing complex to complementary messenger RNA for degradation or translational repression. In medicine, researchers apply shRNA libraries to screen cancer cells, host-pathogen interactions, drug-response pathways, and therapeutic targets, often linking gene knockdown with changes in survival, proliferation, or signaling. These experiments help validate disease mechanisms and prioritize candidates for drug development, while careful controls address variable knockdown efficiency and off-target effects.

MISSION LentiPlex Pooled shRNA Library Screening in Mammalian Cells

Here we use a human LentiPlex pooled library and traditional sequencing methods to identify gene targets promoting cell survival. We demonstrate how to set up and deconvolute a LentiPlex screen and validate the results.
