Tissue Specific Glucose Uptake

Tissue-specific glucose uptake is the regulated movement of glucose from the bloodstream into distinct tissues, where it supports energy production, storage, and biosynthesis. Uptake depends on tissue-specific glucose transporters and hormonal signals: insulin promotes GLUT4 movement to the cell surface in skeletal muscle and adipose tissue, while the liver, brain, and other tissues use different transporters and regulatory mechanisms. Measuring these patterns helps explain how glucose is distributed after meals and how tissues maintain energy balance. In medicine, tissue-specific glucose uptake is central to understanding insulin resistance, diabetes, metabolic disease, and the development of imaging-based diagnostics and targeted therapies.

In this video, we demonstrate an ex vivo technique to measure extracellular glucose analog depletion in harvested mouse organs. It involves incubating the mouse tissue in a solution containing fluorescently-labeled deoxyglucose, which is taken up by the cells. The glucose analog uptake can be indirectly determined by measuring the fluorescence of the incubated medium.
