Tumor Dna Isolation

Tumor DNA isolation is a laboratory method for extracting genomic DNA from tumor cells or tissue so that cancer-associated genetic changes can be studied. The process typically disrupts cells and nuclei, releases DNA, removes proteins and other contaminants, and purifies the nucleic acid for downstream analysis; careful handling helps preserve DNA quality and reduce interference from degraded or non-tumor material. In medicine, isolated tumor DNA supports polymerase chain reaction, sequencing, copy-number analysis, and biomarker testing, helping researchers and clinicians characterize mutations, compare tumor profiles, and inform disease classification or treatment research. Its reliability directly affects the accuracy and interpretability of molecular cancer studies.

This video describes DNA isolation protocol from stool samples of colon tumor-bearing mice. The extracted DNA is amplified using PCR, and the product is further cleaned using a magnetic beads-based approach. This method can be used to determine the effect of environmental enrichment on colon microbiota and animal mortality.

DNA isolation protocols can be fast and straightforward or complex and time-consuming depending on the type and quality of DNA required for further processing. For example, plasmid DNA extraction is a bit more complicated than genomic DNA extraction because of the need for an appropriate lysis method to separate plasmid DNA from gDNA during isolation. However, for specific applications, such as long-range DNA sequencing that require a good yield of high- quality DNA samples, we need to follow...

Side Population Assay: A Method to Isolate Side Population of Tumor Cells from Zebrafish

2023

The following protocol describes a method for isolating the side population of T-cell Acute Lymphoblastic Leukemia (T-ALL) tumor cells, consisting of stem cells, from an adult Zebrafish. The side population lacks prominent cell markers and hence we use Hoechst 33342 dye efflux assay to isolate them.
