Tumor Propagation

Tumor propagation is the growth and spread of abnormal cells that sustain a neoplasm within a tissue or across the body, making it central to cancer progression and treatment. It involves repeated cell proliferation, survival under changing conditions, invasion through surrounding extracellular matrix, entry into blood or lymphatic vessels, and, in metastatic disease, exit from circulation and colonization of distant organs; genetic and epigenetic changes can support these behaviors. Studying tumor propagation helps researchers model disease progression, identify biomarkers, evaluate therapies, and understand why some tumors recur or resist treatment.

Isolation and Propagation of Circulating Tumor Cells from a Mouse Cancer Model

Circulating tumor cells (CTCs) have been shown to play an important role in tumor metastasis. Here, a method for the isolation and propagation of CTCs from the whole blood of a syngeneic mouse tumor model of hepatocellular carcinoma (HCC) metastasis is described.

Establishment and Propagation of Human Retinoblastoma Tumors in Immune Deficient Mice

A method is described to propagate human retinoblastoma tumors in mice. Tumor cells are directly injected into the eyes of immune deficient mice. Secondary tumors have been successfully established using both cells directly harvested from human tumors and cultured tumorspheres.

Limiting dilution cell transplantation assays are used to determine the frequency of tumor-propagating cells. This protocol describes a method for generating syngeneic zebrafish that develop fluorescently-labeled leukemia and details how to isolate and transplant these leukemia cells at limiting dilution into the peritoneal cavity of adult zebrafish.
