Agarose Embedded Slice

Agarose embedded slice is a tissue-preparation method in which nervous tissue is enclosed in a supportive agarose matrix before sectioning, helping produce stable slices for microscopic and experimental analysis. After the agarose solidifies around the specimen, a vibratome or similar instrument advances a blade through the matrix and tissue under controlled conditions, reducing movement and deformation during cutting. In neuroscience, this approach supports the preparation of brain and spinal cord sections for neuroanatomical mapping, immunohistochemistry, and cellular morphology studies. By improving section integrity and handling, agarose embedding facilitates consistent examination of neural circuits, tissue organization, and disease-related structural changes.

We have developed a tissue-processing technique utilizing a vibratome and agarose-embedded lung tissue to generate lung sections, thereby permitting the acquisition of high-resolution images of lung architecture. We employed immunofluorescence staining to observe spatial protein expression using specific lung structural markers.
