Astrocyte Maturation

Astrocyte maturation is the developmental process through which immature glial cells acquire the morphology, molecular identity, and physiological functions of mature astrocytes, which are essential for healthy neural circuits. During this process, astrocyte precursors differentiate and respond to intrinsic developmental programs and signals from neurons and the surrounding tissue, increasing their capacity for neurotransmitter uptake, ion buffering, metabolic support, and regulation of synapses. Understanding these changes helps researchers distinguish immature from mature astrocytes in developing brain tissue and cell culture. Maturation models support neuroscience studies of brain development, neurodegeneration, injury, and disease, while improving the design of human neural models and potential cell-based therapies.

The video demonstrates a simple method to culture murine astrocytes in isolation from other brain cells. During culturing, by using astrocyte-specific media and inhibiting the adherence of non-astrocytic cells, the method selectively cultures astrocytes. These cultured astrocytes exhibit stellate morphology with fine processes.
