Axon Initial Segment

The axon initial segment (AIS) is a specialized region at the junction between a neuron’s soma and axon that typically initiates action potentials, making it central to neuronal communication. Its dense concentration of voltage-gated sodium and potassium channels, organized by ankyrin-G and associated cytoskeletal proteins, enables rapid membrane depolarization when synaptic inputs bring the membrane to threshold. The AIS also helps regulate neuronal excitability by controlling action-potential initiation and filtering signals arriving from dendrites and the cell body. Studying its molecular organization and structural plasticity supports research on neural circuits, sensory processing, development, and disorders involving abnormal neuronal firing.

An in vivo imaging protocol to monitor primary sensory axons following dorsal root crush is described. The procedures utilize wide-field fluorescence microscopy and thy1-YFP transgenic mice, and permit repeated imaging of axon regeneration over 4 cm in the PNS and axon interactions with the interface of the CNS.
