Histological Immunohistochemical Analyses

Histological immunohistochemical analyses are techniques that detect and localize specific proteins within tissue sections, linking cellular identity and structure to biological function. The method uses antibodies that bind target antigens, followed by enzyme-linked or fluorescent detection that produces a visible signal under a microscope. In neuroscience, researchers apply these analyses to map neurotransmitters, receptors, neuronal markers, and disease-associated proteins across brain regions. Comparing staining patterns can reveal cell types, tissue organization, molecular changes, and responses to injury or treatment, making immunohistochemistry valuable for studying neural circuits, development, neurodegeneration, and the effects of experimental interventions.

Here we describe a protocol for multiplex fluorescent immunohistochemical staining and imaging for the simultaneous localization of multiple cancer-associated antigens in lymphoma. This protocol can be extended to the colocalization analysis of biomarkers within all tissue sections.

Consecutive cryo-sections are collected to enable histological applications and enrichment of RNA for gene expression measurements using adjacent regions from a single mouse skeletal muscle. High-quality RNA is obtained from 20 - 30 mg of pooled cryosections and measurements are directly compared across applications.

Histological Staining of Neural Tissue

In order to examine the cellular, structural and molecular layout of tissues and organs, researchers use a method known as histological staining. In this technique, a tissue of interest is preserved using chemical fixatives and sectioned, or cut into very thin slices. A variety of staining techniques are then applied to provide contrast to the visually uniform sections. In the study of neuroanatomy, histological techniques are frequently applied to visualize and study nervous system tissue.
