Brain Specific Delivery

Brain-specific delivery is the design of therapeutic systems that direct drugs, nucleic acids, or diagnostic agents to the brain while limiting exposure in other tissues. It addresses the blood-brain barrier, whose tight endothelial junctions restrict movement from circulation into neural tissue. Delivery systems may use receptor-mediated transcytosis, nanoscale carriers, intranasal transport, or localized administration to improve passage, release, and cellular targeting. In neuroscience, these approaches support treatment of brain tumors, neurodegenerative disease, infections, and genetic disorders, while enabling more precise imaging and reducing systemic toxicity.

Convection-Enhanced Delivery of Antibodies into a Mouse Brain

2025

This video demonstrates a convection-enhanced delivery (CED) technique for antibody delivery into the mouse brain. The skull of an anesthetized mouse is exposed, and a burr hole is created at the target area for antibody delivery. A step catheter containing antibodies is inserted at the target site, and via an infusion pump, a pressure gradient is created to deliver antibodies at a controlled rate.

Intranasal Delivery of siRNA to the Mouse Brain Using a Positioning Device

Source: Ullah, I., et.al. A Positioning Device for the Placement of Mice During Intranasal siRNA Delivery to the Central Nervous System. J. Vis. Exp. (2019)This video demonstrates a precise intranasal administration technique for delivering neuron-targeting peptide-siRNA complexes to the mouse brain using a specialized positioning device. Anesthetized mice are secured in a head-down-and-forward position to optimize siRNA inhalation while preventing lung drainage. The targeting peptide...

Begin with a harvested rat brain submerged in artificial cerebrospinal fluid in a petri dish.Deposit the magnetic millirobot onto the brain cortex. The millirobot, termed MANiAC, consists of magnetically aligned nickel nanorods embedded within a microcapsule composed of the biocompatible hydrogel alginate and loaded with a dye as a model drug.Position an external magnetic field generator beneath the petri dish to generate a uniform rotating magnetic field. The nickel nanorods align and rotate...

Delivery of a Traumatic Brain Injury to a Mouse Brain

This video demonstrates the delivery of Traumatic Brain Injury (TBI) in mice. After exposing the skull, the TBI site is marked. The mouse is then positioned under the TBI device. The impact is delivered by lifting the metal rod and allowing it to drop freely onto the impactor, which is in contact with the skull.

MicroRNAs play crucial roles in the brain and are potential targets for modeling neuro-degeneration. However, perturbing miRNA levels is challenging due to the short length of miRNA and inaccessibility of the brain tissue. This video presents a method for antagomir design and brain specific delivery using a neuropeptide in mice.
