Brain Volume Measurement

Brain volume measurement is the quantitative assessment of the size of the whole brain or specific structures, providing an important marker of anatomy and change in neuroscience. Researchers typically acquire structural magnetic resonance images, distinguish brain tissue from surrounding fluid and skull, and use image segmentation to delineate regions whose volumes are calculated from their constituent voxels. These measurements support studies of development, aging, neurodegenerative disease, injury, and treatment response by revealing patterns of atrophy or abnormal growth. Careful image quality control, consistent processing, and appropriate comparison with reference populations improve the reliability and interpretability of volumetric results.

Here we present a protocol to simply and reliably measure the lung pressure-volume curve in mice, showing that it is sufficiently sensitive to detect phenotypic parenchymal changes in two common lung pathologies, pulmonary fibrosis and emphysema. This protocol describes real-time iontophoresis, a method that measures physical parameters of the extracellular space (ECS) of living brains.
