Chemical Stimulus Preparation

Chemical stimulus preparation is the systematic process of formulating and handling chemical agents used to activate, inhibit, or otherwise probe nervous system responses. It requires selecting appropriate compounds, preparing defined concentrations in compatible solvents or physiological media, controlling factors such as pH and osmolarity, and delivering stimuli consistently while including suitable vehicle or baseline controls. In neuroscience, carefully prepared chemical stimuli support experiments on neuronal excitability, synaptic signaling, sensory transduction, and behavior. Standardized preparation improves reproducibility, helps distinguish compound-specific effects from handling artifacts, and enables reliable comparisons across cells, tissues, animal models, and experimental conditions.

Inducing Axonal Varicosities with a Micromechanical Stimulus on Neurons

This video demonstrates the induction of axonal varicosities by applying micromechanical stimulus on neurons. The system comprises a micropipette attached to a syringe containing buffer via tubing. Using a micromanipulator, the micropipette is positioned above the neurons. Fluid pressure is then applied to induce varicosities in axons.

Here, we present a protocol on how to determine the quantity and distribution of metals in a sample using synchrotron X-ray fluorescence. We focus on adherent cells, and describe the chemical fixation method to prepare this sample. We then describe how to mount and image the sample using synchrotron X-rays.
